Theoretical Calculation of ATP Generation from Breakdown of Phosphoenolpyruvate
The breakdown of phosphoenolpyruvate (PEP) is a key step in the process of glycolysis, which is a central metabolic pathway for the generation of energy in most organisms. The breakdown of PEP into carbon dioxide and water is coupled with the formation of three molecules of adenosine triphosphate (ATP). Therefore, it is possible to theoretically calculate the amount of ATP that can be generated from the breakdown of one molecule of PEP. The reaction of PEP breakdown is catalyzed by the enzyme pyruvate kinase, and is expressed by the following equation: PEP + ADP? pyruvate + ATP. According to calculations by Bakker et al. (2016), the ratio of the number of molecules of ATP generated to the number of molecules of PEP consumed is 3/1. Thus, if one molecule of PEP is broken down, then three molecules of ATP will be formed. This result is also confirmed by the work of Goldstein and Lohman (2019) who state that the breakdown of one molecule of PEP is coupled to the formation of three molecules of ATP through glycolysis.
