Glucose 6-phosphate (G6P) plays an essential role in regulating hexokinase (HK) enzyme activity in various cell types, including breast tissue. It has been demonstrated that, in breast tissue, G6P stimulates the activity of hexokinase which in turn increases glucose uptake and utilization in the glycolytic pathway (Razzaque, 2020). This, in turn, leads to the generation of ATP, a key energy source for cells. G6P also serves as a regulator of hexokinase expression and activity, as it increases the rate of hexokinase transcription and translation (O’Neill, 2016). However, this normal effect of G6P on hexokinase activity may not occur in tumor cells. This is because tumor cells typically have a higher level of glycolysis than normal cells, which may lead to an increased concentration of G6P in tumor cells than in non-cancerous tissues (Ng et al., 2021). If this is the case, the G6P may not be able to activate hexokinase effectively and thus, the enzyme activity may not be significantly increased.
