Enzyme substrate binding is a crucial part of many biochemical processes, yet it can be difficult to achieve. In order for a substrate to bind to the enzyme’s active site, several forces and factors must be overcome. These include electrostatic interactions, van der Waals forces, hydrophobic interactions, and steric hindrance. Electrostatic interactions refer to forces between charged atoms. When a substrate molecule is introduced to an enzyme’s active site, the electrostatic forces between the substrate and the enzyme must be overcome in order for the substrate to bind. This interaction can be modulated by the pH of the solution, as well as by the presence of cofactors or other molecules that may increase the affinity of the enzyme for the substrate (Boucif et al., 2016). Van der Waals forces refer to the attractive forces between the electron clouds of two molecules. These forces are generally weak and can be overcome with proper positioning of the substrate molecule in an enzyme’s active site. Hydrophobic interactions refer to the interactions between nonpolar molecules and water molecules.
