The principle behind the Next Generation Sequencing (NGS)

The principle behind the Next Generation Sequencing (NGS) methodology of Illumina Sequencing is to use the principle of bridge amplification to generate short DNA fragments (150-300 base pairs) which are then sequenced (Mallick et al., 2017). The process begins by ligation of short fragments of DNA to adapters, which are then separated into individual clusters and added to a flow cell. These DNA fragments are then amplified to create millions of copies of the original fragments. The fragments are then sequenced by adding fluorescently labeled nucleotides that can be detected and recorded by the Illumina sequencer (Singh et al., 2020). The sequencing process is repeated millions of times, producing millions of reads which are then aligned to a reference genome to generate a sequence. The end result is a high-quality, massive amount of data which can be used for a variety of applications such as variant detection, gene expression analysis, and microbial identification (DeJesus et al., 2019).
